Crateva tapia L. bark lectin (CrataBL) shows low acute systemic toxicity with mild tissue responses

Resumen

CrataBL, una lectina bifuncional e inhibidor de proteasas aislada de la corteza de Crateva tapia L., presenta actividades anticoagulante, antiinflamatoria, antimicrobiana y antitumoral. Sin embargo, su perfil toxicológico sigue estando poco caracterizado. Este estudio evaluó la toxicidad aguda de CrataBL en ratones Swiss hembra tras una única administración por vía oral o intraperitoneal a una dosis de 200 mg/kg. Los animales fueron monitoreados durante 14 días para evaluar la mortalidad, los signos clínicos y conductuales, el peso corporal y el consumo de alimento y agua. También se evaluaron los pesos de los órganos, los parámetros hematológicos y bioquímicos, así como las alteraciones histopatológicas. CrataBL no indujo mortalidad ni signos observables de toxicidad, independientemente de la vía de administración. El aumento de peso corporal, el consumo de agua, los pesos de los órganos y los marcadores bioquímicos séricos de la función hepática y renal permanecieron sin cambios en comparación con los controles, aunque se observó una ligera reducción en el consumo de alimento tras la administración oral. Los parámetros hematológicos se mantuvieron, en su mayoría, sin alteraciones, observándose únicamente cambios menores en las poblaciones de leucocitos. El análisis histopatológico reveló alteraciones leves en el hígado, los riñones y los pulmones, incluyendo infiltrados inflamatorios focales, cambios estructurales renales, degeneración glomerular, engrosamiento de la pared alveolar y placas hialinas intraalveolares, mientras que el bazo y el corazón permanecieron sin alteraciones. En conjunto, CrataBL presentó una baja toxicidad aguda, lo que respalda un perfil preliminar de seguridad favorable. Se requieren estudios adicionales para establecer su caracterización toxicológica a largo plazo y su aplicabilidad terapéutica.
