Carapichea ipecacuanha is a medicinal plant of pharmacological importance, and in vitro cultivation represents an alternative for its propagation and conservation. This study aimed to evaluate the effects of different LED light wavelengths on the in vitro development and multiplication of two C. ipecacuanha accessions. Explants were inoculated onto MS medium supplemented with sucrose, IBA, and Phytagel and exposed to five light spectra (white, green, blue, yellow, and red). Accessions 590 and 836 were evaluated using a completely randomized design in a 5 × 2 factorial arrangement with three replicates. After 180 days of cultivation, plantlet height and shoot number were evaluated, along with chlorophyll content and genetic characterization based on the ribosomal DNA ITS region. For plantlet height, accession 836 showed the highest mean under green light, whereas accession 590 showed the greatest growth under blue light. For shoot number, accession 590 showed the highest mean under blue light, while accession 836 showed the highest means under green and red light. However, differences among light treatments were not statistically significant, indicating only trends in the responses of the accessions to different light qualities. Accession 836 exhibited higher chlorophyll a and b contents under yellow light, whereas accession 590 showed higher concentrations under white light. In both accessions, chlorophyll b contents were higher than chlorophyll a contents. Molecular characterization revealed high genetic similarity between accessions 590 and 836, which were grouped within the same haplotype (H22), together with sequences from the Amazon region. The different LED light spectra promoted distinct morphological and physiological responses between the accessions, although no statistically significant differences were detected. Molecular analysis confirmed the genetic similarity between the accessions and their grouping within the same Amazonian ITS haplotype.
